The overall view is that while your paper will be of great interest to the field it is not one of the most competitive in terms of general interest. If you have confidence to submit this leading journal, please remember that it is a leading journal, and you have to maximize general interest. So that your paper will have a better chance to get approved lying on the solid foundation of your article. Of course, you could encounter a competitor who is in the same field and his paper is more excellent than yours. Roll a dice. There is nothing you could do. If you want to submit it to Science, you must be excellent and lucky.

Switching journals does not necessarily mean getting reviewed. There have been cases where articles spent over half a month at both S and N journals and passed the advisor stage, but were rejected for review and given a transfer option. Surprisingly, when they were submitted to SA and NC, they were rejected immediately.
